Scholarship Description: This project will test the hypothesis that in plant cells Golgi bodies are generated de novo at export sites on the ER and that the numbers of Golgi stacks in the cell are regulated by division of export sites and re-absorption of Golgi components along the retrograde pathway back into the ER. Alongside this it will also test the hypothesis that Golgi-associated proteins âÂ" the golgins âÂ" are responsible for organising the 3-D structure of the ER-Golgi interface and developing cisternal stacks. The majority of the molecular players at the plant ER exit sites have now been identified and cloned, including key exit site components Sec16 and Sec12, Sar1, the COPII coat components (Sec 23/24, 13/31), the key SNARES and a number of golgins or matrix/putative tethering proteins. All of these clones are available to us and many were made in our lab. The project will involve the use of high end microscopy instrumentation including live cell imaging, optical tweezers and serial block face imaging SEM for 3-D cellular reconstruction.
